Interest in two-dimensional HPLC (2DLC) in pharmaceutical analysis has been rapidly growing in recent years, as evidenced by recent journal publications and presentations at analytical conferences. Although the technology and theory of two-dimensional separations were introduced more than 30 years ago, 2DLC applications of liquid chromatography have mainly been used in proteomics, natural product, biological sample and polymer analysis and have been limited to pharmaceutical analysis until recently. The increased interest is likely due to new demands in pharmaceuticals and recent advances in 2DLC hardware, software and column technology. This article describes a hardware and software system based on the Sintecon HPLC 10.600 chromatograph for analyzing samples with complex matrices in 2D liquid chromatography mode. The advantages of the Sintecon chromatograph compared to other commercially available systems are listed.
